چکیده:

Summary Extraction of Taxol (paclitaxel) from hazel (Corylus avellane L.) cell cultures have been widely demonstrated, though with limited yield. Nanoparticles have been recently fascinating due to their unique properties to penetrate cells and expressing elicitation effects. This study evaluates the role of aluminum oxide (Al2O3) and tungsten trioxide (WO3) nanoparticles (NPs) on the production of bioactive compounds in leaf derived callus cultures of C. avellane. To obtain in vitro seedlings seeds of C. avellana were used as a source of in vitro seedling plants. The seeds of this species have low potential germination rate, so the cold conditions at 4 °C in the darkness were used to breaks seed dormancy, for three days. Seeds grown in the control treatment were kept at room temperature. The germinated seeds were collected and re cultured onto MS medium supplemented with different types of plant growth regulators (PGR) for regeneration and establishment the proliferation stage. The regeneration treatment contained 2 mg.L -1 BA, 0.1 mg.L -1 GA3 and 0.01 mg.L -1 IBA. The calli were obtained from seedling in Murashige and Skoog medium supplemented with varying concentrations of auxin (NAA) and cytokinin (Kin), and were sub-cultured every 21 days. After getting a line, the calli were treated with different concentrations (0, 2.5, 5, 10 mg. L − 1 ) of Al2O3NPs and WO3NPs at their logarithmic growth phase. Changes in total phenolic and flavonoid content, fatty acid profiles, and Taxol production were assessed. Paclitaxel was extracted and quantified via HPLC, and gene expression levels of key biosynthetic enzymes i.e., Taxadiene synthase (TS), Deacetyl baccatin III-10-O acetyltransferase (DBAT), and phenylalanine aminomutase (PAM) were analyzed using RT-qPCR.
